羟基喜树碱对乳腺癌细胞MCF-7细胞信号转导相关基因的影响

摘  要:
目的:从全基因组角度探索羟基喜树碱对乳腺癌细胞MCF-7细胞信号转导相关基因的影响,寻找羟基喜树碱在乳腺癌治疗中潜在的新靶点.方法:HCPT诱导MCF-7细胞,MTT法检测HCPT对细胞增殖抑制.选择40mg/mL的HCPT作为诱导剂量组,并设置阴性对照,进行基因组芯片杂交实验.结果:HCPT对MCF-7细胞的增殖抑制作用均呈浓度依赖性,即随着HCPT浓度的增高,MCF-7的增殖受到明显抑制.HCPT诱导后,共获得了表达下调基因46个,表达上调基因9个,其中insulin-like growth factor binding protein1(IGFBP1)是表达下调(-4.01031)最大的信号转导基因,表达上调最大的信号转导相关基因是一广谱表达基因hemoglobin2(HBG2)(+3.804527),它在癌症发生中的作用未知.Q-PCR实验结果与芯片结果具有良好的一致性.结论:HCPT诱导乳腺癌细胞增殖抑制,可能涉及多个基因.

摘  要:
To explore the potential target of medication of breast carcinoma,whole genome microarray analysis was done by comparing the cells induced by HCPT with the control group. MTT was used to detect whether the proliferation of tumor breast cell line MCF-7 was inhibited and find the proper dosage of HCPT for induction. Whole genome microarray was use to search the differentially expressed genes with function of signal transduction. Proliferation of the MCF-7 cells was reduced with increasing HCPT concentration. Fifty genes were found to be differentially expressed in MCF-7 cells,of which 46 genes showed inhibited expression and 9 indicated elevated expression,when induced by HCPT. Expression of insulin-like growth factor binding protein1 (IFGBP1) (-4.010 31) gene was inhibited most,while hemoglobin2 (HBG2) (+3.804 527) without known function in tumoresis was elevated most. Reliability of the microarray result was validated by Q-PCR by choosing two differentially expressed genes.
