反溶剂-高压均质法制备羟基喜树碱纳米粒工艺及其性质研究

摘  要:
为了减小羟基喜树碱的粒径,该研究首先采用了反溶剂法(以二甲基甲酰胺,三氯甲烷为反溶剂)对羟基喜树碱原粉微粉化,使其粒径降低至1 000 nm以下,再通过高压均质法将羟基喜树碱微粉纳米化.在反溶剂过程中,考察了多种溶剂及反溶剂,确定了以二甲基甲酰胺为溶剂,三氯甲烷为反溶剂;在高压均质过程中,考察了药物浓度、均质压力及均质循环数,最终确定了制备羟基喜树碱纳米粒的最优条件为:药物浓度为0.8 mg/mL,均质压力为60 MPa,均质循环数为20个循环.以上条件制备的羟基喜树碱纳米粒粒径为135.1 nm;通过激光粒度分析仪及X射线衍射(XRD)对羟基喜树碱原粉与纳米粒进行了性质分析,与原粉相比羟基喜树碱纳米粒的粒径及结晶度均有所降低.

摘  要:
To decrease the particle size of hydroxycamptothecin,the nanoparticle of hydroxycamptothecin in an antisolvent process(using dimethylformamide as solvent and chloroform as atsolvent)and a high pressure homogenization technique was studied in the research.In antisolvent process,we inspected multiple solvents and antisolvents,then chose dimethylformamide and chloroform.In the high pressure homogenization process,drug concentration,pressure and the number of cycles were investigated.And we prepared the nano hydroxycamptothecin with the particle size of 135.1 nm under the optimal condition(drug concentration 0.8 mg/mL,60 MPa and 20 cycles).To detect the characterization of raw hydroxycamptothecin and nano hydroxycamptothecin,we adopted laser particle analyzer and differential scanning calorimetry(DSC).The particle size and crystallinity of nano hydroxycamptothecin decreased greatly compared with raw hydroxycamptothecin.
