四种化学还原法制备胶体金的比较研究

摘  要:
以柠檬酸三钠、鞣酸、抗坏血酸、硼氢化钠为还原剂,采用4种不同化学还原法制备出单分散性好的胶体金样品;比较研究了4种方法制备的胶体金的光学性质、贮存稳定性及其对蛋白质的标记能力。结果表明,不同方法制备得到的胶体金样品其外观色泽均呈现红色略带紫色或葡萄酒红色,可见光谱图最大吸收峰波长为510~525 nm,胶体金粒径在5~20 nm之间。不同贮存条件对胶体金稳定性有影响,其中以4℃条件下贮存的胶体金最稳定;不同方法制备得到的胶体金对蛋白质的最佳标记量有影响。

摘  要:
In this paper,colloidal gold was prepared by four chemical reduction methods with citrate sodium,tannic acid,ascorbic acid and sodium borohydride as reductants.The optical properties,storage stability and labeling protein ability of colloidal gold prepared by the four methods were compared.The results showed that the color of all the colloidal gold was red with a little purple or red wine color.The maximum absorption wavelength was 510~525 nm.Particle diameter of colloidal gold was 5~20 nm.Stability of colloidal gold was affected by different storage conditions.Colloidal gold remained optimal stability at 4℃.Labeling protein amount of colloidal gold was affected by different preparing methods.
