提高昆虫杆状病毒表达系统重组蛋白表达水平的技术

摘  要:
昆虫杆状病毒表达系统已广泛用于各种重组蛋白的生产。然而,由于各种因素重组蛋白的表达量远远达不到理想的水平。提高重组蛋白的表达水平成为提高昆虫杆状病毒表达系统效率的核心问题。根据目前的研究,提高外源基因表达水平的技术主要包括抑制目的蛋白降解、合理调控目的基因的转录、优化启动子类型以及利用活体昆虫而非培养细胞,如用家蚕幼虫或蛹进行表达等。这些技术也为家蚕成为更加高效的生物反应器奠定了基础。

摘  要:
Insect Baculovirus Expression Vector System (BEVS) has been widely used in a variety of recombinant protein production.However, the expression of foreign protein is limited due to various factors.High-level expression of recombinant proteins becomes the core issue of improving the efficiency of BEVS.So far, the main techniques for improving the expression of foreign gene included the inhibition of protein degradation, positive regulation of foreign gene transcription, optimization of promoters and utilization of live insect rather than cell lines, such as silkworm larva or pupa as host.These techniques also lay a firm foundation for developing silkworm as efficient bio-reactor.
