鹿角盘蛋白质酶法水解作用研究

摘  要:
研究鹿角盘中蛋白质酶法水解条件。用胰酶、胰蛋白酶、木瓜蛋白酶在相同条件下水解鹿角盘蛋白质,比较氨基氮生成率,选择适宜酶类。用所选定的酶在不同温度(T)、不同底物浓度比(E/S)和不同水解时间(t)水解鹿角盘蛋白质,以氨基氮生成率为指标确定最佳水解条件。胰蛋白酶为适宜酶,最佳水解条件为:温度45℃,pH 8.0,底物浓度7.5%,酶与底物浓度比6 000μ/g,时间6h。胰蛋白酶是水解鹿角盘蛋白质较适宜的酶,在上述最佳条件下,水解鹿角盘蛋白质,氨基氮生成率达42%,氮溶解指数达93%,水解液中蛋白质绝大多数为低肽分子,更有利于机体消化吸收。

摘  要:
To study protein enzymatic hydrolysis conditions from antler plate.Antler plate protein was hydrolyzed with trypsin,pancreatin and papain under the same condition,amino nitrogen production rate was compared,suitable enzyme was choosed.The protein enzymatic hydrolysis in antler plate was researched with different temperature,enzyme/substrate ratio,and hydrolysis time.The optimum parameters of reaction were determined with amino nitrogen as the index.Trypsin is suitable for protein hydrolysis in antler plate.The results showed that the optimum conditions of hydrolysis are the temperature at 45℃,the original pH 8.0,substrate concentration 7.5%,enzyme/substrate ratio(E/S)6 000μ/g,and hydrolysis time 6h.Trypsin is suitable enzyme for protein hydrolysis in antler plate.Under optimum conditions,amino nitrogen production rate is up to 42%,the index of soluble nitrogen is up to 93% in hydrolysate.The most of protein in hydrolysate is low molecular weight peptide,which is good for digestion and absorption of human body.
