α——淀粉酶同工酶在大麦发芽过程中的变化

摘  要:
苏啤1号大麦芽水浸提液,用等电聚焦分离α—淀粉酶,淀粉胶板检测,可得到约19条同工酶带。其中等电点在4.7、5.9、6.1—6.7的八条酶带活性强。等电点在4.7的酶,活性从发芽的第四天起检测到,第五、六、七天增强;等电点在5.9的酶,发芽第一天就检测到活性,在第七天增强;等电点在6.1—6.7的酶,发芽第一天就检测到活性,第二天就急剧增强活性;等电点在4.7—5.9之间的九条酶带,发芽过程中活性几乎不变。0.2ppm赤霉素溶液处理大麦,只能使发芽过程中,α—淀粉酶的含量增多,不能改变α—淀粉酶同工酶等电聚焦酶谱。对日本大麦(二棱)甘木二条、南斯拉夫293号(二棱)进行了研究,发现和苏啤1号的酶谱不同。但赤霉素的作用是一致的。

摘  要:
Su Pi No.1 malted barley extracted in water,a-amylase separated by iselectriciocusing ( IEF ) and datermina ted by starch-gel plate,had obstained about 19 bands of the isoenzymos. 8 Bands of the isoenzymes ( PI4.7,5.9,6.1-6.7) had strongly activity,a-Amylas eisoenzymes s ( PI,4.7 ) had ac tivity in the fourth day of germinating and activity wa increasing in the fifth,sixth,seventh day; a-amylase isoenzymes ( PI5.9 ) had activity in initail stage of germination and had strongly activity in the seventh day of germination; a-Amylase isoenzymes ( PI,6.1-8.7) had activity in inntail stage of germination.Activity of 9 bands of the isoenzymes ( PI,4.7-5.9) changed hardly during germination. Su Pi No.1 barley was incubated in 0.2ppm gibberelin slotion which only increased amount of a-amylas and did not change a-amylase isoenzymes zymogram of IEF. Investigation for GanMu ErTiao ( two row,japan),XanSiLaFu o.293 discovered that thy zymog erms of then were different from the zymogratrn of SuPiNo.1.But,action of gibberelin was consistent.
