基于ITS序列对3个侧耳菌种的分子鉴定

摘  要:
对实验室长期保存的3个侧耳(Pleurotus)菌种核糖体DNA的ITS(internal transcribed space)序列进行了PCR扩增,克隆测序后,与从GenBank中获得的相关序列一起构建了侧耳属系统发育树。结果显示,Pleurotus djamor CBS665.97与肺形侧耳(Pleurotus pulmonarus)的距离最近,Pleurotus ostreatusCBS596.96与红侧耳(Pleurotus djamor)的距离最近,Pleurotus dryinus ACCC51152与盖囊侧耳(Pleuro-tus cystidiosus)的距离最近。因此得出这三个菌种为命名错误,Pleurotus djamor CBS665.97应为Pleu-rotus pulmonarus CBS665.97,Pleurotus ostreatus CBS596.96应为Pleurotus djamor CBS596.96,Pleurotusdryinus ACCC51152应为Pleurotus cystidiosus ACCC51152。

摘  要:
The internal transcribed spaces(ITS) of the three Pleurotus spawns were cloned by PCR and sequenced.An ITS phylogenetic tree was constructed based on combination of sequence data set from GeneBank.The phylogenetic tree showed that Pleurotus djamor CBS665.97 was close to Pleurotus pulmonarus,Pleurotus dryinus CBS596.96 was mostly close to Pleu-rotus djamor,and Pleurotus dryinus ACCC51152 was mostly close to Pleurotus cystidiosus.Therefore,it could be concluded that these Pleurotus spawns’ names were wrong.Pleurotus djamor CBS665.97 should be named as Pleurotus pulmonarus CBS665.97;Pleurotus ostreatus CBS596.96 should be named as Pleurotus djamor CBS596.96;and Pleurotus dryinus AC-CC51152 should be Pleurotus cystidiosus ACCC51152.
