基于线粒体DNA 16S rRNA基因鉴别畜禽肉中鸭源性成分研究

摘  要:
研究以鸭线粒体16S rRNA基因序列为靶位点设计特异性引物,以常见畜禽肉包括牛肉、羊肉、猪肉、兔肉、鸽肉、鹌鹑肉、鸡肉、鸭肉、鹅肉等参考动物肌肉DNA为模板,进行常规PCR和荧光定量PCR扩增,建立鸭源性成分检测方法;并将鸭肉DNA模板浓度进行8个梯度稀释,检测方法灵敏度。结果显示,所设计的引物对能够有效地对鸭源性成分进行快速检测,具有较强的特异性,方法灵敏度较高,可达pg级,可快速准确地检测畜禽肉中鸭源性成分。

摘  要:
To establish a fluorogenic quantitative PCR method,which could identify the components of duck origin,16 S rRNA gene sequence was used as target site,and the specific primers of duck were designed. The DNA of common livestock and poultry meat including beef,mutton,pork,rabbit meat,pigeon meat,quail meat,chicken,duck and goose were used as templates. And duck DNA template was diluted into eight gradients for detecting the sensitivity. The results showed that the designed primer pair had a quite specificity and could detect the duck origin effectively and quickly. And the method had a high sensitivity,which reached pg level,could detect the duck origin in livestock and poultry meat quickly and accurately.
